Bovine embryo induces an anti-inflammatory response in uterine epithelial cells and immune cells in vitro: Possible involvement of interferon tau as an intermediator OAK
Talukder, Anup K.; Yousef, Mohamed S.; Rashid, Mohammad B.; Awai, Kensuke; Acosta, Tomas J.; Shimizu, Takashi; Okuda, Kiyoshi; Shimada, Masayuki; Imakawa, Kazuhiko; Miyamoto, Akio.
Recent observations suggest that the bovine uterus starts to react to the early embryo immediately after its arrival from the oviduct. The present study aimed to investigate the effect of the early developing embryo on the immune-related gene profile in bovine uterine epithelial cells (BUECs) in vitro, and to further examine the impact of conditioned media (CM), either from embryo-BUEC co-culture or embryo culture alone, on gene expression in peripheral blood mononuclear cells (PBMCs). First, BUECs were co-cultured with morulae (n = 10) for D5-D9 (D0 = IVF), and gene expression in BUECs was analyzed. Subsequently, PBMCs were cultured in CM from embryo-BUEC co-culture or D5-D9 embryo culture, and gene expression was evaluated. In BUECs, the embryo induced...

Ca2+ entry, efflux and release in smooth muscle Biol. Res.
MATTHEW,A; SHMYGOL,A; WRAY,SUSAN.
Control of smooth muscle is vital for health. The major route to contraction is a rise in intracellular [Ca2+], determined by the entry and efflux of Ca2+ and release and re-uptake into the sarcoplasmic reticulum (SR). We review these processes in myometrium, to better understand excitation-contraction coupling and develop strategies for preventing problematic labours. The main mechanism of elevating [Ca2+] is voltage-gated L-type channels, due to pacemaker activity, which can be modulated by agonists. The rise of [Ca2+] produces Ca-calmodulin and activates MLCK. This phosphorylates myosin and force results. Without Ca2+ entry uterine contraction fails. The Na/Ca exchanger (NCX) and plasma membrane Ca-ATPase (PMCA) remove Ca2+, with contributions of 30%...

CCR3 antagonist impairs estradiol-induced eosinophil migration to the uterus in ovariectomized mice BJMBR
Araújo,J.M.D.; Silva,L.A.S.; Felix,F.B.; Camargo,E.A.; Grespan,R..
Eosinophils are abundant in the reproductive tract, contributing to the remodeling and successful implantation of the embryo. However, the mechanisms by which eosinophils migrate into the uterus and their relationship to edema are still not entirely clear, since there are a variety of chemotactic factors that can cause migration of these cells. Therefore, to evaluate the role of CCR3 in eosinophil migration, ovariectomized C57BL/6 mice were treated with CCR3 antagonist SB 328437 and 17β-estradiol. The hypothesis that the CCR3 receptor plays an important role in eosinophil migration to the mouse uterus was confirmed, because we observed reduction in eosinophil peroxidase activity in these antagonist-treated uteruses. The antagonist also influenced uterine...

Differences in progesterone concentrations and mRNA expressions of progesterone receptors in bovine endometrial tissue between the uterine horns ipsilateral and contralateral to the corpus luteum OAK
TAKAHASHI, Hiroto; HANEDA, Shingo; KAYANO, Mitsunori; MATSUI, Motozumi; 羽田, 真悟; 茅野, 光範; 松井, 基純.
Because the establishment of pregnancy begins at the uterine horn ipsilateral to the corpus luteum (ipsi-horn) in cattle, levels of progesterone (P4) and receptor expression in the endometrial tissue, which regulate the intrauterine environment for embryo development, may differ between the ipsi-horn and the uterine horn contralateral to corpus luteum (contra-horn). The aim of the present study was to determine the endometrial tissue P4 concentrations and nuclear progesterone receptor (PGR), progesterone receptor membrane component 1 (PGRMC1) and PGRMC2 mRNA expressions in the cranial and middle parts of the uterine horns during the luteal phase. The results showed higher endometrial tissue P4 concentrations in the cranial part of the ipsi-horn than in...

Experimental use of fibrin glue derived from snake venom in non-pregnant canine uterus J. Venom. Anim. Toxins incl. Trop. Dis.
Moraes,J.R.E.; Correia,P.H.A.; Camplesi,A.C.; Moraes,F.R..
This study evaluated the use of fibrin glue derived from snake venom in the healing process after canine hysterorrhaphy. Three groups of four animals were submitted to uterine hysterotomy followed by wound closure. In Group 1, double-layer suture was used, the first with Schimieden pattern, the second with Cushing pattern; in Group 2, only fibrin glue; and in Group 3, the same as for Group 1 but with fibrin glue as suture reinforcement. Results indicated that fibrin glue produced less inflammation in the exudative phase, and exacerbated deposition of connective tissue and angiogenesis in the proliferative and maturation phases of the healing process, favoring its evolution.

Expression of mdr isoforms in mice during estrous cycle and under hormone stimulation Genet. Mol. Biol.
Schiengold,Marion; Schwantes,Lavínia; Ribeiro,Maria F.; Lothhammer,Nívia; Gonzalez,Tatiana P.; Chies,Jose Artur Bogo; Nardi,Nance B..
The multidrug resistance (MDR) phenotype is associated with the expression of P-glycoprotein (Pgp), coded by the multigenic mdr family. Mice present the isoforms mdr1 and mdr3, which are responsible for multidrug resistance, and mdr2, that is involved in the transport of phospholipids. mdr1 expression has more recently been associated also with the secretion of steroid hormones. This work presents an RT-PCR analysis of the expression of mdr isoforms, in several organs of mice during different phases of the estrous cycle. Additionally, females were ovariectomized, submitted to different hormone treatments, and their uterus was analyzed for the expression of mdr isoforms. The results show that in the adrenal gland and ovaries mdr1 is the main isoform during...

Expression of Uterine Sensitization-Associated Gene-1 (USAG-1) in the Mouse Uterus During the Peri-Implantation Period OAK
MAEDA, Kanami; LEE, Dong-Soo; YANAGIMOTO UETA, Yoshiko; SUZUKI, Hiroshi; 鈴木, 宏志.
Rat uterine sensitization-associated gene-1 (USAG-1) mRNA is expressed in the uterus during the peri-implantation period, and its mRNA expression in uterine epithelial cells is highest on day 5 of pregnancy. On the other hand, since changes in USAG-1 mRNA expression in the mouse uterus are not seen during the estrous cycle, USAG-1 expression might be specifically regulated by embryonic factors rather than by the maternal environment. However, the expression pattern and function of USAG-1 in the mouse uterus have not been determined. Thus, we examined the tissue-specific USAG-1 mRNA expression in the uteri of ICR mice during peri-implantation using real-time quantitative PCR. Uterine tissues, such as the myometrium, luminal epithelium, and stroma, were...

Histological Remarks of the Uterus of Sympterygia acuta (Garman, 1877) and Sympterygia bonapartii (Müller & Henle, 1841) (Chondrichthyes; Rajidae) International Journal of Morphology
Díaz-Andrade,María Constanza; Lopez-Cazorla,Andrea; Galíndez,Elena Juana.
Chondrichthyes constitute a conserve and vulnerable clade of increasing importance. However, there is scarce knowledge about the morphological aspects of the reproduction, so that only one work exists about the structure of the gravid uterus of an oviparous species. The work presented here describes the morphological, histological and histochemical remarks of the uterus of two Rajids of the Sympterygia genus along the life cycle. Both species presented paired uteri that lead independently to a common urogenital sinus. The uteri of immature females are filiform, translucent and unspecialized tubes. When sexual development starts, all the genital system displays a transitional and progressive change. In all studied females, independently of the maturing...

Intra-uterine experimental infection by Ureaplasma diversum induces TNF-α mediated womb inflammation in mice Anais da ABC (AABC)
Silva,Jamile R.; Ferreira,Lício F.A.A.; Oliveira,Percíllia V.S.; Nunes,Ivanéia V.; Pereira,Ítalo S.; Timenetsky,Jorge; Marques,Lucas M.; Figueiredo,Tiana B.; Silva,Robson A.A..
Ureaplasma diversum is an opportunistic pathogen associated with uterine inflammation, impaired embryo implantation, infertility, abortions, premature birth of calves and neonatal pneumonia in cattle. It has been suggested that the intra-uterine infection by Ureaplasma diversum can cause vascular changes that hinder the success of pregnancy. Thus, the aim of this study was to evaluate the changes of intrauterine site of A/J mice in estrus or proestrus phase inoculated with Ureaplasma diversum. The infection was monitored at 24, 48 and 72 hours by the PCR methodology to detect the Ureaplasma in the inoculation site and the profile of circulating blood cells. Morphological changes, intensity of inflammation and the production of cytokines were compared. The...

Morphological Changes in the Pregnant Uterus of the Smooth Hound Dogfish Mustelus schmitti Springer, 1939 (Gatuzo) (Condrichthyes, Triakidae): Microscopic Study and Phylogenetic Reproductive Implications International Journal of Morphology
Galíndez,E. J; Díaz Andrade,M. C; Moya,A. C; Estecondo,S.
Elasmobranchs are important, not only as an economic resource but also for their phylogenetic position. Their slow growth and long reproductive cycles, together with the high fisheries pressure exerted, attempt to the knowledge of all biological parameters of species. In this work we analyze the histology of the pregnant uterus of the gatuzo, a viviparous aplacental shark. The mucosa depicts modifications similar to those of placental species and others proper of histotrophism. Likewise, a network-like muscular layer in the mucosa was described for first time in the genus. The presence of vitelline plates in the gut of at term embryos suggests that the maternal supplies are not enough for the big fetus development. The relationships between maternal...
